WV 66 will be covered in bicyclists that day. I'd avoid it.
The rest of your route is fine. A bit zig-zaggy yet not the worst route.
Here are some alternatives:
I81-I70-I68 West to US219 South in Maryland. Travel through Deep Creek, Oakland, Parsons, Elkins, Huttonsville on the way to Marlinton. Lots of ups and downs, good places for gas rests and food, scenery.
I81 south to Lexington, VA then VA/WV 39 through Goshen Pass into Marlinton. (Secret Road).
I81 to I66 at Strasburg -- take VA/WV 55 all the way to Seneca Rocks, then WV 28/92 to Marlinton. After entering WV on Hwy 55, stay on the old road while everybody else uses the 4 lane...
I81 to Woodstock, VA; VA 42 south to either US 33 or US 250 into WV; then WV 28/92 again.
Them's the best alternatives fer a city slicker!

That's the route I'm planning on taking and hopefulyl meeting up with Vann around lunchtime (12:30~1:00) at Seneca Rocks. But take note - The interstate section of Mike's directions are reversed. I wanted to post this in time before everyone heads out. We need to do a group picture before this thing is over and I suggest Saturday evening before dinner in front to the Inn. There is room in front for all the bikes to park with the Inn in the background.

Figure casual clothing. I usually wear a suit and tie, but this year I will wear real tree camo, and maybe my t shirt from the Irish Camel Bar in Park City, UT.
You could also wear camo, but the real tree lets you blend in better.
BTW, I am also planning on Seneca Rocks for lunch on Friday. I will get an early start and try US 250, or WV 20, who knows.

Wayne, I think there will be space for you. I am leaving tomorrow morning and the Watoga Super' I emailed didn't expect a busy weekend. If they're out of sites and if they allow it, you can share a site with me.
Funny how the weather report changes! Last night NOAA showed clear and cool all four days...tonight its rain for the first two and all have highs around 60.
Normally I take VA39 that direction but I81 to I64 to 219 this time, coming back Sunday on 39.

Find Huntersville, WV on WV 39 about 6 miles east of Marlinton. Take County Road 21 (big brown state park sign indicates turn) 9-10 miles south.
At sign showing turn for Watoga Park, continue straight south for one more mile to Beaver Creek Campground.
From US219 (about 8-9 miles south of Marlinton) turn onto County Road 27 (signs for Watoga show this) and proceed through Seebert, across bridge and into park. Travel all the way through the park and exit onto County Road 21, turn south one mile, etc.

My bike is loaded on the truck and I'm looking at 501 miles for 9 hours. I'm bringing Tools, a cut open stock muffler, my homemade oil manometer, a mercury carb styx and a 1150R cylinderhead with 2 exhaust valves - - - - for show-n-tell and maybe a few TB synchs.
After driving Ann's 2004 Civic 1,170 miles to Boston MA for a wedding last Saturday, a 9hr, 500 mile Nissan pickup trip is gonna be easy.
I'll be there Thursday evening well before dark.
